Nothing has been revealed about the past of Al Mackenzie. It is known that he was the CIA liaison to the law enforcement agency, S.H.I.E.L.D. prior to the Deltite affair. The Deltites were artificially created duplicates of human beings who took control of S.H.I.E.L.D. After being rescued from the Deltite replicas by Nick Fury, Mackenzie joined the small band of S.H.I.E.L.D. agents who struggled against the Deltite takeover.

After the defeat of the Deltites, Mackenzie remained with Fury as he traveled around the world, closing and dismantling former S.H.I.E.L.D. bases. During this time, he became romantically involved with the Contessa Valentina Allegra de la Fontaine, Nick Fury's long-time girlfriend. This led to an estrangement between Fury and Mackenzie, and Mackenzie and the Contessa returned to the CIA.

Recently, after a brief stint as an agent for the CIA, Mackenzie has joined the new S.H.I.E.L.D. organization created by Fury, and now he often participates in their activities.
